We have developed a cell-culture technique that allows the study of trivalent cationic lanthanide transport with the potential to be an important tool for elucidating the transport pathways of these medically important radionuclides. V79 cells were cultured in either supplemented L15 medium or standard MEM medium, and bound [Ce3+] and [Nd3+] were measured. Since the lanthanides all transport with the anion Cl-, and [Ce3+] and [Nd3+] are a function of the net Cl- flux, the cells were studied for their Cl- fluxes when K+ was substituted for Na+ in the medium. The substitution of K+ for Na+ was also studied for the transport of [Tb3+] because Tb3+ forms stable ternary complexes with Eu3+ and Cl-. A standard L15 cell-culture medium (containing 2 mmol/L KCl) was used. The total cell number, plating efficiency, and colony forming efficiency were determined. V79 cells cultured in medium with K+ were identical to those cultured with Na+. The cell number was unaffected by either [Nd3+] or [Tb3+] exposure (0-200 mumol/L). The plating efficiency was not affected by either [Nd3+] or [Tb3+] exposure (0-200 mumol/L). The colony forming efficiency was decreased with [Nd3+] (0-100 mumol/L) and increased with [Tb3+] (0-200 mumol/L). [Tb3+] uptake was not dependent on [Nd3+] exposure. Although there was a concentration-dependent increase in the intracellular [Ce3+] and [Nd3+] from 200 to 300 mumol/L with [Nd3+], the intracellular [Tb3+] was unaffected by [Nd3+] (100 mumol/L).
